10x chemistries - v2 and v3 3', and 5'?

Hello, thanks for your hard work on Irescue!

I have been asked to run this on a dataset with a mix of chemistries. I am wondering, will Irescue work differently on 10X 3' V2 vs V3 chemistry? What about 10X 5' (5 prime) sequencing?

Hi, you can run IRescue on any 10X single cell/nuclei RNA-seq chemistry, as long as barcodes and UMIs are annotated in the BAM file (by CellRanger, STARsolo or similars).
Some chemistries, especially 3' vs 5'-end, capture different library compositions, so you can expect different results.

To further expand: IRescue is also compatible with SMART-seq and other full-length protocols that do not use UMIs.
