Three FDA-approved disease-modifying therapies — a paradigm shift in rare disease:
- Nusinersen (Spinraza, Biogen/Ionis): antisense oligonucleotide that promotes SMN2 exon-7 inclusion, increasing functional SMN protein. Intrathecal administration every 4 months.
